Hematopoietic Stem Cells represent a very rare population of adult bone marrow cells that have the unique capability of generating the entire blood and immune system for an organism throughtout its lifespan. Over the past few years, there has been enourmous interest in this population of cells, which can be attributed, in part, to the rapidly growing clinical fields of gene therapy and bone marrow transplantation. The increase in the number of studies related to stem cells has spawned a large number of in vitro and in vivo assays designed to characterize purified stem cell populations from both mice and men.

To date, there has been no collection of these assays in a single volume that succinctly described the inherent value and limitation of each experimental approach. This volume enables both basic scientists and clinical researchers to isolate and functionally characterize purified stem cells from various sources, to genetically modify stem cells using viral gene delivery systems, and to describe various molecular analyses that can be performed iwth a small number of cells.


Forty-one international specialists contribute 20 chapters to this volume, providing a variety of key laboratory methods for investigating hematopoietic stem cells (HSC). The text includes approaches using mouse and human tissues of both embryonic and adult origin, methods for the purification and genetic modification of stem cells, stem cell expansion protocols, and in vitro and in vivo assays, with techniques for studying a variety of stem cell topics, including blood formation, blood-based disease, and blood cancers such as leukemia.
